Nicotine Affects Behaviour, Morphology and Cortical Cytoskeleton of Amoeba proteus

چکیده
Nicotine, a natural alkaloid of tobacco leaves and roots, is the main factor for human tobacco addiction. To assess its toxic effect on motile cells we have made series of experiments on free-living Amoeba proteus. It was found that nicotine changed cell morphology, inhibited locomotion, as well it led to degradation of actin cytoskeleton and finally to cell destruction. Obtained results suggest necrosis not the apoptosis as the mechanism of cell death, however many phenomena concerning structure of cytoskeleton and physical cell disintegration suggest that process might to be active.

Extracellular Calcium Changes the Morphology of Induced Pinocytosis in Amoeba proteus
In Amoeba proteus a typical induced pinocytosis is manifested after external application of different agents: aminoacides, proteins, monovalent cations. This pinocytosis is a kind of fluid-phase endocytosis driven by cortical cytoskeleton and may be compared to macropinocytosis in Dictyostelium discoideum in which large fluid-filled vesicles are formed (Maniak 2002). During induced pinocytosis ...
متن کاملEffects of Blocking of Endogenous RHO Family GTP-Binding Proteins on Morphology, Adhesion and Locomotion of Amoeba Proteus.
Rho family GTP-binding proteins are known to control cellular processess associated with actin-based cytoskeleton such as cell migration, cytokinesis, endocytosis and exocytosis or muscle contraction [1]. While there is a number of data on the role of these proteins in higher Eukaryota, the studies on protozoans are practically limited to Dictyostelium that does not contain Rho-like and Cdc42-l...
متن کاملFilamentous Structures in Amoeba Proteus Visualized in Scanning Microscope.
The dynamic cytoskeleton in Amoeba proteus is a filamentous gel network, composed mainly of F-actin microfilaments accompanied by actin binding proteins, distributed in the peripheral cortex beneath the cell membrane and around the nucleus. As well the cell shape changes as the motility primarily depend on the activity of these structures. In contrast, the endoplasm of amoeba is commonly consid...
متن کاملNucleus-associated actin in Amoeba proteus.
The presence, spatial distribution and forms of intranuclear and nucleus-associated cytoplasmic actin were studied in Amoeba proteus with immunocytochemical approaches. Labeling with different anti-actin antibodies and staining with TRITC-phalloidin and fluorescent deoxyribonuclease I were used. We showed that actin is abundant within the nucleus as well as in the cytoplasm of A. proteus cells....
